Well, that's a very big question without a simple answer.

 

It all depends very much on exactly what you want to do.

 

A good start is here:

This tells you what everything means in the css and how to change ot to change colours, fonts etc.

 

For more drastic changes you will need to do a little research, depending on what you want to do. Probably 99% of what you want to do has already been done and there will be threads about it so try the search and the other wiki docs, both links in my sig.

 

There is a LOT of info in the wiki docs so go there first.

And, browse the "my store" section of this forum, to see what people are doing. Take a look at my site- I changed the overall look to some extant, I had to hard code each page for quite a lot of it- becaue I didn't understand css at the time

You can try one of the template systems in the contributions section. I haven't had any luck with any of them, yet, myself, but plenty of people are doing fine with them!
