Check in your site's global configuration - if you have enabled gzip compression, turn it off and see if this makes a difference. So where did mainfile.php come from?

You (or your ISP) are using URL rewriting, but it doesn't fit well with your current PHP-Nuke code. Maybe you installed, say, Nuke Sentinel, and you forgot to make the necessary changes in the code - something like that, I guess. I think mainfile.php is from phpuke, so you should ask on that forum.
