chmod doesnot work!!!

I am working on a new store. I had it working for a couple of days. All of a sudden ...BOOOM!!! I cannot get it to chmod the configure.php now. I tried it thru ftp...webftp...and PHP. I also tried 777,644,444,400 with no luck. I have another store working on the same host. Any thoughts?
